Crucell Developing West Nile Vaccine, Kimron Licence Pact
AMSTERDAM -(Dow Jones)- Dutch vaccine and antibody company Crucell N.V. said Wednesday that it is developing a human West Nile vaccine based on the company's proprietary PER.C6 cell line.
West Nile virus is present in birds and is spread to humans by mosquitoes. In man, about 1% of infected individuals contract a severe, sometimes fatal brain infection.
The virus has been transmitted between humans through blood donations, organ transplants and breastfeeding. Since 1999, West Nile virus is spreading throughout the United States of America and Canada. There are currently no vaccines to protect humans against infection.
Crucell conducted pre-clinical studies in a geese animal model with an experimental vaccine, demonstrating disease free survival after a lethal challenge with the West Nile virus. Geese are susceptible to, and usually do not survive, West Nile virus infection. Geese therefore represent a good and stringent model for testing experimental vaccines.
Crucell also announced a license agreement with Kimron Veterinary Institute of Israel. The program will focus on the development and registration of a whole killed West Nile veterinary vaccine based on Crucell's PER.C6 cell line technology.
The Kimron Veterinary Institute is the diagnostic and research arm of the Veterinary Services of the Israeli Ministry of Agriculture.
Under the terms of the agreement, Kimron will pay Crucell a license fee, milestones and a royalty on sales of the vaccine in Israel. Crucell will receive the commercial rights to the whole killed West Nile veterinary vaccine outside of Israel and will pay Kimron a royalty on sales.
Kimron has previously developed and registered a whole killed veterinary West Nile vaccine produced on mouse brains. It currently manufactures and distributes 800,000 doses of this vaccine per annum.
Over the last six months, Kimron and Crucell have conducted experiments with a PER.C6(TM)-based whole killed West Nile vaccine in geese. Based on the results of these tests, Kimron intends to replace their existing West Nile veterinary vaccine with the PER.C6-based vaccine.
The new vaccine is a "whole killed" vaccine, which means that the virus will be inactivated, yet be able to elicit a strong and effective immune response. The new vaccine is also expected to have improved purity at competitive costs. Kimron expects to register the PER.C6 West Nile veterinary vaccine in 2004.
In Israel, West Nile virus affects the country's geese population, resulting in severe morbidity and mortality. The virus is commonly carried by both domesticated and wild birds and is sprea d to humansby mosquitoes. The PER.C6- based veterinary vaccine will besuitable for geese, horses and potentially other animals.

West Nile Virus
The West Nile virus appears to be firmly established in the United States, and researchers expect its continued spread and entrenchment in wildlife populations. Since 1999, WNV has been detected in humans, horses, birds, or mosquitoes in 44 states and the District of Columbia. Only Hawai'i, Alaska, Oregon, Nevada, Utah, and Arizona have yet to report detection of WNV.
According to the U.S. Centers for Disease Control and Prevention, 4156 people in the the U.S. tested positive for the West Nile Virus in 2002, and 284 people died of the virus. Many more people were likely to have been infected with the virus, but experienced mild or no symptoms, and were never tested.
Statistically, a person's risk of contracting West Nile is low. In most areas where the virus is established, only 1% of the area's mosquitoes carry the virus. Less than 1% of people bitten by these infected mosquitoes develop serious complications from the virus; the remainder exhibit flu-like symptoms, or no symptoms at all. Those at highest risk are the elderly and people with weakened immune systems; it is important, however, for all people to protect themselves from mosquito bites to minimize the risk of infection.

Leiden, The Netherlands, June 11, 2003 - Dutch vaccine and antibody company Crucell N.V. (Euronext, NASDAQ: CRXL) announced today that it is developing a human West Nile vaccine based on the company's proprietary PER.C6(TM) cell line.
"The pre-clinical studies were very encouraging. We demonstrated that a PER.C6(TM)-based vaccine protects against the Israel 1998 Goose strain of West Nile virus," said Dr. Jaap Goudsmit, Crucell's Chief Scientific Officer. "The fact that this strain is closely related to the NY99 strain, which caused the West Nile outbreak in the USA, triggered our decision to invest in development of a West Nile vaccine for humans. Crucell intends to announce clinical development timelines later this year."

Electron micrograph of the Ebola virus (CDC)
A vaccine against the deadly Ebola virus has been found to protect monkeys with a single dose, offering a new way to stop the fast-acting disease, U.S. researchers have announced.
The vaccine was made using a new approach that should work against a range of other viruses - technology that may offer a quick way to develop an instant vaccine against new infections, such as SARS, or even a biological weapon, the scientists report in today's issue of the journal, Nature.
Dr Gary Nabel, the top vaccine expert at the U.S. National Institute of Allergy and Infectious Diseases and leader of the study, said he was surprised a single shot worked. "If we can do this for Ebola virus, I am hopeful we can apply it to other infectious outbreaks," he said.
Ebola, a newly-emerged and frightening virus that kills anywhere between 50% and 90% of victims though massive internal bleeding, has resisted vaccine efforts so far.
Nabel and colleagues started with a common virus called an adenovirus - the culprit behind many cases of the common cold as well as some more serious infections. They added one piece of DNA from the Ebola virus, a glycoprotein.
Traditional vaccines use a whole virus - live, killed or weakened - to help the immune system recognise the invader. The DNA vaccine relies on a strong immune response to the adenovirus, and the immune system recognises the Ebola virus simply by recognising the glycoprotein.
To the researchers' surprise, monkeys given a single jab with this vaccine were 100% protected from what should have been a deadly dose of Ebola, they report in this week's issue of the journal Nature.
Years of failure
"After years of developing candidate Ebola vaccines that protected rodents but failed in primates, it is gratifying to have a vaccine that holds great promise for protection of humans," said Dr Peter Jahrling, a senior research scientist at the U.S. Army Research Institute of Infectious Diseases, who also worked on the study.
In April, an outbreak of Ebola in the Congo Republic killed more than 120 people as well as scores of gorillas living in a nearby wildlife preserve. A vaccine would be used to control such outbreaks in both people and animals, Nabel said.
"'Ring vaccination' might be used to stop the spread of the Ebola virus during acute outbreaks, just as this strategy was used to contain smallpox in the past," Nabel said.
In ring vaccination, everyone who has been in contact with a patient, as well as all members of the patient's household, are vaccinated. The new vaccine is also safer than the smallpox vaccine, which uses a live virus related to smallpox.
"This begins to change our minds about how vaccines can be used," Nabel said. Instead of vaccinating large populations against diseases just in case, such a vaccine could be used tactically, when an outbreak starts, to control it quickly.
Nabel said the approach may be used in the development of vaccine against SARS (Severe Acute Respiratory Syndrome). The newly-emerged disease, which befell 8,437 people around the world and killed more than 800, seems under control now but experts suspect it may reemerge during the northern winter flu season.
The National Institute of Allergy and Infectious Diseases is working with Maryland-based GenVec on vaccines against both SARS and the AIDS virus. For the Ebola virus, The institute is working with Dutch biotechnology company Crucell NV. They hope to test it in humans soon.
The U.S. National Institute of Allergy and Infectious Diseases is still pursuing an older approach to an Ebola vaccine that it has been working on with Merck and Co Inc, called a DNA prime-boost, which may offer longer-term protection against the virus.

Crucell N.V.: Ebola Vaccine Holds Promise for Protection in Humans
In an article published in Nature(1) this week, test results have shown that a new Ebola vaccine gives macaque monkeys 100% protection from the Ebola virus with a single dose. Scientists at the Vaccine Research Center (VRC), part of the National Institute of Allergy and Infectious Diseases (NIAID), of the National Institutes of Health (NIH), and scientists from the United States Army Medical Research Institute of Infectious Diseases (USAMRIID) carried out the research. The vaccine is a prototype adenoviral vaccine, which is made by inserting a portion of Ebola virus DNA into an adenovirus, the virus that causes the common cold. The results suggest that humans may be protected by a single vaccine injection.
Crucell entered into a Cooperative Research and Development Agreement with the NIH in 2002 to jointly develop an Ebola vaccine for human use. Currently Crucell is producing the experimental vaccine for testing in human trials on its proprietary PER.C6(TM) production platform. Crucell's PER.C6(TM) technology makes large-scale production of the vaccine possible.

Crucell and GeneMax Enter PER.C6 License Agreement in Gene Delivery Field to Advance Clinical Trial
LEIDEN, Netherlands, August 12, 2003 (PRIMEZONE) -- Dutch biotechnology company Crucell N.V. (Euronext:CRXL) (NasdaqNM:CRXL - News) and US-based GeneMax Corp. (OTC BB:GMXX.OB - News) (Frankfurt:GX1) today announced that they have entered a license agreement for Crucell's PER.C6(TM) cell line technology. Under the terms of the agreement, GeneMax will use Crucell's PER.C6(tm) technology for research in the field of adenovirus-based gene delivery. GeneMax has also obtained an option for a non-exclusive commercial license to use PER.C6(tm) cells to manufacture and sell products in the field of gene delivery. Crucell will receive upfront and annual payments for the research license. If the research license is converted into a commercial license, Crucell will receive additional annual payments and royalties on future sales of PER.C6(TM)-derived products. Further financial details were not disclosed.
This license represents an important building block in our program to move GeneMax's cancer vaccine into clinical trials. We perceive the Crucell vector to be the best available for delivering the TAP gene,'' said Ronald Handford, President & CEO of GeneMax.
We signed the first PER.C6(tm) license in 1998. Since that time PER.C6(tm) has become the 'industry standard' in the area of adenovirus-based gene delivery . Currently, six PER.C6(tm)-based products are in clinical trials,'' commented Dinko Valerio, Crucell's President and CEO. As interest in gene delivery rebounds, we believe new opportunities for future agreements will further strengthen PER.C6(tm)'s value to this industry.''

Crucell and Kimron Veterinary Institute Announce Anticipated Approval of West Nile Veterinary Vaccine Early 2004
LEIDEN, The Netherlands, Sept. 03, 2003 (PRIMEZONE) -- Dutch biotechnology company Crucell N.V. (Euronext:CRXL) (NasdaqNM:CRXL - News) and Israeli Kimron Veterinary Institute announced today that they anticipate approval of a veterinary West Nile vaccine in early 2004. Kimron had previously expected to register the PER.C6(TM)-based West Nile veterinary vaccine at the end of 2004. Based on encouraging results from the initial pre-clinical studies, Kimron now will conduct a field trial during the 2003 mosquito season to ensure that the licensed vaccine is available before the start of the 2004 mosquito season. Kimron signed a license agreement with Crucell in June 2003 to use Crucell's PER.C6(TM) cell line technology to develop a West Nile veterinary vaccine. The PER.C6(TM)-based West Nile veterinary vaccine is a whole-killed vaccine, which means that the virus is inactivated, yet able to elicit a strong and effective immune response. The new vaccine is expected to have improved purity at competitive costs.
Kimron will commence a large trial involving approximately 2,000 geese to test the PER.C6(TM)-based West Nile veterinary vaccine under field conditions. The geese will be vaccinated two times within a 14-day interval followed by direct exposure to the live West Nile virus. Depending on the success of the field trial, it is likely that the PER.C6(TM)-based West Nile veterinary vaccine will be approved for use in geese in early 2004.
Geese are natural targets for the West Nile virus. Geese can contract a West Nile virus infection resulting in West Nile fever and possibly neurological disease and death. In Israel, West Nile virus affected the country's goose population resulting in severe morbidity and mortality until vaccination with a West Nile veterinary vaccine was implemented. Kimron intends to replace their existing West Nile veterinary vaccine, which is currently produced on mouse brains, with the PER.C6(TM)-based vaccine.
Kimron has commercial rights to the whole-killed West Nile veterinary vaccine in Israel, and will pay Crucell a royalty on sales. Crucell will receive the commercial rights to the vaccine outside of Israel.
Human Vaccine Against West Nile Virus In a separate program, Crucell is also developing a West Nile vaccine based on the whole-killed virus specifically for use in humans. As geese are naturally susceptible to West Nile infection they represent a good and stringent model for testing experimental vaccines. Crucell intends to announce clinical development timelines for the human West Nile vaccine later this year.
Overview of West Nile Virus
West Nile virus is a member of the flavivirus family, which includes yellow fever and St Louis encephalitis and is commonly found in Africa, West Asia and the Middle East. West Nile virus is carried by birds and is spread to humans by mosquitoes. In humans, infection with West Nile virus usually results in a case of West Nile fever, which is a mild disease characterized by flu-like symptoms. In about 1% of cases it causes severe and sometimes fatal inflammation of the brain and surrounding membranes (meningitis, encephalitis or meningoencephalitis). Currently, there is no vaccine available to protect humans against infection with West Nile virus

Crucell Announces 2003 Annual Results
Successful Transition to Product Focused Company
LEIDEN, The Netherlands, Jan. 26, 2004 (PRIMEZONE) -- Dutch biotechnology company Crucell N.V. (Euronext:CRXL) (NasdaqNM:CRXL - News) today announced its 2003 financial results. Crucell's revenues for the year ended December 31, 2003 were 7.4 million (US$ 9.3 million) compared to 9.6 million (US$ 12.0 million) for 2002. A significant up-front fee from a commercial contract signed in December 2003 is not included in revenue, but is deferred and will be recognized as revenue in future years. Total deferred revenues at December 31, 2003 were 13.8 million (US$ 17.4 million) compared to 6.0 million (US$ 7.6 million) for 2002.
The net loss in 2003 was 23.4 million (US$ 29.4 million), compared to 55.7 million (US$ 70.0 million) in 2002, which included a goodwill impairment charge of 30.9 million (US$ 38.9 million). In 2003, research and development expenses and selling, general and administrative expenses were below 2002 expense levels. Cash used during the year amounted to 23.4 million (US$ 29.5 million) compared to 9.6 million (US$ 12.1 million) in 2002. At December 31, 2003 cash and cash equivalents stood at 87.2 million (US$ 109.7 million).
Chief Financial Officer Leon Kruimer commented: ``The Company has successfully made the transition from a research and technology-focused organization to an organization fully dedicated to bringing innovative products to the market. Our recent agreement with Aventis Pasteur to jointly develop influenza vaccines frees up additional resources for our other in-house programs. During the last year we have increased our investment in preclinical studies across our core programs, and are now gearing up to enter the clinic.''

LEIDEN, Netherlands, April 14, 2004 (PRIMEZONE) -- Dutch biotechnology company Crucell N.V. (Euronext:CRXL) (NasdaqNM:CRXL - News) today announced its financial results for the first quarter of 2004. Crucell's revenues for the first quarter were 4.0 million (US$ 4.9 million) compared to 2.1 million (US$ 2.6 million) for the same quarter in 2003. The increase relates primarily to revenues recognized from an agreement with Aventis Pasteur for the co-development of a new generation of influenza vaccines, as well as to additional licensing and other revenues. The Company reported a net loss for the quarter of 7.0 million (US$ 8.5 million) compared to 3.6 million (US$ 4.4 million) in the same quarter last year. The increase is primarily due to non-cash expenses related to long-term incentive plans. Cash and cash equivalents increased by 1.0 million (US$ 1.2 million) to 88.2 million (US$ 107.4 million) during the quarter.
``Cash burn is the most important financial indicator by which we manage Crucell,'' said Chief Financial Officer Leon Kruimer. ``We met our objectives to increase revenues in the first quarter and despite the fact that we acquired ChromaGenics for 4.0 million in cash, we increased our cash reserves.'' Chief Executive Officer Ronald Brus added that ``Crucell is committed to increasing revenue, yet at the same time we continue to invest significantly in development of our vaccines. Our development programs are now our top priority and we maintain our guidance for a cash burn of 20 to 25 million for 2004 as our programs accelerate throughout the year.''

schöne Charts hast du da.....leider hab ichs zu späte gesehen......... :(

CRXL Analysts Day Webcast Nov. 16, 2004
Notes by Trudy the Monkey. Images by Crucell from the webcast itself.
Open with CEO Ronald Brus
2004 has been a successful year; A year that brought us closer to the heart of what our mission is all about: To be the leading biotech company developing vaccines and antibodies to prevent or treat infectious diseases.
We will continue to leverage PER.C6, but will more and more focus on the development of vaccines and antibodies out of the Crucell shop.
We are going to announce Today a new antibody for the disease rabies.
In 2004:
We met all our research and development milestones.
We made progress in all vaccine programs.
We found new partners: NIH, IAVI, and AERAS.
We advanced protein production, and will receive an important DSM milestone payment.
Today we can announce permission from AME/Eli Lilly to say that a monoclonal antibody related to inflammation, based on PER.C6, with blockbuster potential, has entered clinical studies in the US.
In 2004, we gained 17 new PER.C6 licences.
Focusing on our own products, in fields in which Crucell can make a difference:
We believe, in the field of influenza ($1.5 billion+ market), we can change the way vaccines are produced.
We believe we can be the first to produce a safe, efficacious West Nile vaccine ($400 million+ market).
For Ebola ($100 million+ market) and Malaria ($200 million+ market), we think we can demonstrate unrivaled efficacy and safety.
We picked rabies ($100 million+ market) because of its similarity to the flu area. We believe we can come up with a monoclonal antibody that will be the treatment of choice for this disease.
It is no longer bold to state that our technology is behind every major vaccine initiative:
Malaria (w/GSK, SAIC)
Aids (w/Merck, IAVI)
Tuberculosis (w/AERAS)
Pandemic Influenza (w/Aventis Pasteur)
Endemic Influenza (w/Aventis Pasteur)
Ebola (w/NIH)
Maximum effort to have PER.C6 widely endorsed by industry. We're very keen to create this critical mass of companies behind our technology. There are 300+ products produced now with the help of the PER.C6 cell line technology.
Which brings us to our business model...
Now, with all these partners, and the revenues that are coming in, we can finance our programs from different sources, and reduce the overall investment that Crucell has to make.
Already twice this year, due to the significant increase in revenues, we had to downward revise burn projections, etc. FOLLOW the URL.....

Also sag mal € 80 Million/Jahr.
Meine Gedanke dabei:
"I have great expectations, Mr Pip......"
Also gestern war dann Backtesting.
BTW: Due Your Own Due Diligence....
Hourly Chart:
Also dann fragt mann sich ab, konnte sowas miet CRXL passieren.
Edwards ist eine US MidCap Company, also netter Vertrag.
URL: http://www.crucell.nl
Kurs: € 8.62 d.h. +2.6 %
Umsatz: 249 k
Just my 2 cents...
Aufgepasst !
€ 12.5
Point&Figure Kursziel ist: $ 20.2
Chart: courtesy StockCharts.com
Just my 2 cents
Darum ist meine Position: Long.

gute Analyse und gut eigekauft,
bis zum nächsten Mal, Gute nacht
Gestern war der sogenannter "Day of the Share" in Amsterdam.
Vielleicht "a storm in a teacup" ?

URL: http://www.behr.nl/Beurs/aex2005.html
Schlusskurs: $ 12.50 - also wieder ein neues Jahreshoch.
Just my 2 cents....
Charts: courtesy chartnet.nl
http://phx.corporate-ir.net/phoenix.zhtml?c=124404&p=IROL-NRText&t=NewsRelease&id=654955&highlight=
Zitat: "Dutch biotechnology company Crucell N.V. (Euronext, NASDAQ: CRXL) announced today that Merck & Co., Inc. (NYSE: MRK) has met a clinical development milestone in connection with the PER.C6® technology licensed to Merck for its investigational HIV vaccine program."
Im Pre-market ~6 % UP.
"I have great expectations, Mr Pip...."

Crucell's Cell-Culture Technology to Be Part of Major U.S. Government Influenza Pandemic Vaccine Program
Leiden, The Netherlands, April 1, 2005 -Dutch biotechnology company Crucell N.V. (Euronext, NASDAQ: CRXL) announced today that its partner sanofi pasteur, the vaccines business of the sanofi-aventis Group, has been awarded a $97 million contract by the U.S. Department of Health and Human Services (HHS). The objective of the contract is to accelerate the licensure in the U.S. of a PER.C6® based cell-culture influenza vaccine and vaccine manufacturing facility. Crucell will be a subcontractor for the program. Terms of the subcontracting agreement between Crucell and Sanofi pasteur were not disclosed.
The project is part of the U.S. government's effort to increase influenza vaccine manufacturing capacity in the event of a pandemic or other influenza health emergency.
As a result of the agreement, sanofi pasteur has announced that it will accelerate its PER.C6® influenza vaccine program. At the end of three years, Phase 1 and 2 clinical studies will be completed and Phase 3 will be underway. In addition, sanofi pasteur announced it will deliver to the HHS a feasibility plan for the construction of a U.S.-based and licensed cell-culture production plant for supplying up to 300 million monovalent influenza vaccine doses annually.Under the terms of the contract, sanofi pasteur will accelerate the existing project in order to complete a detailed design approximately two years earlier than originally intended. Sanofi pasteur will entirely refit a vaccine development facility for the project and design and test the process from small to mid-sized industrial scale. Lonza Biologics plc., a biotechnology contract manufacturer and subsidiary of The Lonza Group, will contribute to key activities related to the scale up to very large volumes of PER.C6® cell culture and engineering design of the production facility. The contract does not include the actual construction of the plant.
Und noch ein Vertrag fuer Crucell...
__________________________________________________ _____
Reuters: AMSTERDAM, April 14 (Reuters) - Dutch biotechnology firm Crucell (CRCL.AS: Quote, Profile, Research) (CRXL.O: Quote, Profile, Research) said on Thursday it has signed a contract with the U.S. Vaccine Research Center to manufacture vaccines against Ebola, boosting its shares.
Crucell said it would receive 21.4 million euros ($27.43 million) to produce up to 10 batches of its Ebola vaccine based on its PER.C6 gene technology to be used in clinical studies in humans.
Crucell's share price, which had already more than doubled this year before Thursday's announcement, was up 3.5 percent at 11.78 euros at 0743 GMT, after earlier touching 11.95 euros, making it one of the top gainers on the Amsterdam bourse. Crucell's PER.C6 gene technology uses human cells as a platform to produce drugs. Researchers are also using the technology in hopes of developing vaccines against illnesses such as HIV and cancer.
"This news confirms our positive view on the company," Rabo Securities, which rates Crucell "outperform", said in a note. "Positive newsflow on the vaccine development programmes should provide further catalysts for the share in 2005."
Earlier this month, Crucell said its partner Sanofi Pasteur had won a $97 million U.S. contract for which Crucell would be a subcontractor. Analysts estimated this would yield the Dutch firm 30 million to 40 million euros over the next three years.
Crucell and Sanofi Pasteur, the vaccine unit of drug maker Sanofi Aventis (SASY.PA: Quote, Profile, Research) (SNY.N: Quote, Profile, Research) , will make an influenza vaccine based on Crucell's technology and will speed up licensing.
Analysts noted at that time that the U.S. Department of Health and Human Services had not yet granted contracts to other influenza vaccine makers, giving a strong indication of the competitive advantages of Crucell's technology.

Jedoch: Caveat emptor - DYODueDiligence...
Als Alternative dieser Breakout
Hi P
TIA
Gestern gab Fortune (Magazin) eine LIste von "25 Breakout Companies" fuer 2005. Dabei war auch Crucell erwaehnt. Unter diese companies wird verstanden:
We picked the brains of gurus and geeks to come up with our 13th annual list of upstarts who are changing the game. Today's esprit de cool runs from San Diego to Beijing and includes everything from gadgets to biotech.

Human Monoclonal Antibody Against West Nile Virus Shows Promise in Animal Studies
Leiden, The Netherlands, June 10, 2005 - Dutch biotechnology company Crucell N.V. (Euronext, NASDAQ: CRXL) today announces the discovery of a monoclonal antibody for protection against West Nile virus. Study results have demonstrated that the fully human monoclonal antibody, derived from patients who survived West Nile infection, effectively neutralized the virus in cell culture and fully protected mice from an otherwise lethal infection.........

Crucell
Technology: Cells for vaccine production
Sector: Biotech
Headquarters: Leiden, Holland
Website: www.crucell.com
If the deadly bird flu simmering in Southeast Asia boils up into a pandemic, demand for influenza vaccines will explode. So will Crucell's stock price. The company offers a line of human cells that's emerging as a key ingredient for making the vaccines. Currently vaccine production entails growing flu viruses in fertilized chicken eggs; the viruses are extracted, deactivated, and processed into flu shots. The eggs come from high-tech poultry farms that would be hard to expand fast. Boosting vaccine production with Crucell's methods would be relatively easy—its cells, in which flu viruses thrive, are kept in big vats called bioreactors that can be readily multiplied to expand capacity.
Recently the U.S. government awarded $97 million to Sanofi Pasteur, the vaccine unit of Sanofi Aventis, to fund its transition to Crucell technology for making flu vaccines in the U.S. The process is also being used to develop vaccines for malaria, Ebola virus, and West Nile virus. Crucell is hawking its technology to make many other bioengineered medicines too. A raft of drugmakers, including GlaxoSmithKline and Roche, have licensed its "PER.C6" cells for that purpose. If enough customers follow suit, Crucell's investors won't have to wait for a flu pandemic to see the stock take off. — David Stipp

The new cell-based vaccine will employ culture techniques licensed from the Dutch company Crucell N.V. It will shorten the production process by several months and allow banking frozen cells for later use. Phase 1 and 2 clinical studies (safety and dose range finding studies, tests of efficacy) will be followed within three years with Phase 3 trials (larger scale pre-marketing tests of safety and efficacy). Sanofi-Pasteur will also design a full-scale manufacturing process for large quantities of vaccine by refitting an existing facility and provide engineering plans for the US construction of a cell-culture vaccine manufacturing facility to supply up to 300 million doses of a monovalent (single strain) influenza vaccine. The contract, however, does not cover constructing such a facility. (Infection Control)
Sanofi-Pasteur will use Crucell's PER.C6(R) cell-line to grow cells in a nutritive medium in bottles or bioreactors rather than eggs. Usually the cells have been "immortalized" so the culture line can grow indefinitely. Here's how the company website describes the technology:
Crucell's PER.C6® technology, which is currently licensed to numerous organizations and biopharmaceutical firms, is a production technology that is derived from a single healthy, human retinal cell. The original cell was immortalized using recombinant DNA technology, allowing it to replicate indefinitely. This feature enables the cell line to produce biologics [vaccines and similar products] in sufficient quantities for commercial distribution.
Many of today's vaccines are produced on animal-derived substrates, including fertilized chicken eggs and mouse brains. To overcome limitations in production capacity, processing time, and potential safety risks associated with the use of animal-derived substrates, Crucell's PER.C6® technology is an attractive alternative production technology for the manufacturing of inactivated whole virus, live-attenuated, live-vector and subunit vaccines.
PER.C6® technology supports the growth of a wide variety of human disease causing viruses that can subsequently be processed into vaccines suitable for administration to humans. Many viruses have been demonstrated to efficiently replicate on PER.C6® cells including influenza, Japanese encephalitis, respiratory syncytial and parainfluenza viruses. Crucell continues to identify additional virus strains that grow efficiently on PER.C6® cells. In addition, Crucell's PER.C6® technology allows for efficient production of recombinant vaccines.

As Western drug companies work on a vaccine for a deadly form of influenza that scientists fear could mutate to pandemic proportions, laboratories around the Asian-Pacific region -- where the flu strain has claimed all its victims so far -- are pressing ahead with vaccines and tests of their own.
About a dozen companies world-wide currently plan to conduct human trials of vaccines against the H5N1 avian-flu virus by year end, said Klaus Stohr, head of the World Health Organization's flu program. For example, Sinovac Biotech Ltd., a small, Beijing-based company whose stock is traded in New York, is co-developing with the Chinese government a human vaccine targeting the virus, which has jumped the species barrier from birds to humans.
Yesterday, Vietnamese scientists successfully tested a vaccine on chickens in preparation for beginning tests on humans soon, according to Nguyen Tran Hien, director of the National Institute of Hygiene and Epidemiology in Hanoi. The announcement came shortly after officials of the WHO voiced concerns that the country's vaccine development strategy could pose a risk to public safety because of the strain the scientists plan to use.
Mr. Hien said the scientists are closely following the WHO's guidelines and that their work poses no threat. Still, even if Vietnam developed its own human vaccine, it is unclear whether the country could produce enough of it to have a substantial effect.
"Research is only a piece of it," Dr. Stohr said. "Not having the production in parallel might lead to a vaccine that is safe but isn't produced on the scale that is necessary."
Meanwhile, Australian scientists announced a 24-hour test for avian flu that can be used on migratory birds. If it proves reliable, the test could be a critical step in stemming the spread of the disease, according to the WHO, though officials of the Geneva organization haven't yet reviewed the test.
In the past few months, international health authorities have issued increasingly urgent statements about H5N1, which has killed 54 people, including 38 in Vietnam and 12 in Thailand, since late 2003, according to the WHO. There is no evidence yet that the virus can pass easily from person to person. But scientists fear that if it does mutate to become readily transmissible among humans, it could cause a pandemic that would kill millions of people.
An effective vaccine, produced soon enough and in sufficient quantity, could stanch a global outbreak if administered strategically. In addition, some governments are administering vaccines to birds, not as a prelude to human testing but to limit the spread of the virus among the fowl.
-------------------------------------------------------------
Sanofi Pasteur, a U.S. unit of Paris-based pharmaceuticals company Sanofi-Aventis SA, is perhaps furthest along in the development of an effective human vaccine against avian flu. The National Institutes of Health in Washington began recruiting more than 400 volunteers in March to test the company's experimental vaccine at sites around the U.S.
--------------------------------------------------------------
H5N1 may have originated in southern China, where the proximity of birds, pigs and other animals to humans has forged a transgenic path for numerous diseases. In humans, bird flu typically begins with a mild fever, which may then subside before spiking, as well as headache and aching joints. Difficulty in breathing ultimately cuts the flow of oxygen to the brain.
In the absence of a human vaccine, health experts say the best defense so far is Tamiflu, a flu medicine made by Roche Holding AG of Switzerland that has the backing of the WHO.
In the area of testing for the disease's presence in birds, the 24-hour Australian test, which was developed by the Department of Primary Industries in the state of Victoria, is certain to draw interest. But its reliability will be key. There are tests that can give results in several hours, but they are often unreliable.
"Speed is one thing, quality is another. The two have to come together," said Dr. Stohr, who was unaware of the Australian effort. "If you had a very rapid test that was also very accurate, that would be phenomenal."

Flu experts gathering in Memphis to discuss vaccine production.
Flu experts from government, industry and scientific circles will gather in Memphis on friday to come up with a new strategy for producing and distributing flu vaccine around the world.
"The conference will address every aspect of the flu vaccine issue, from vaccine shortages and production issues to novel approaches for rapid development and quick response to emerging flu strains," says Malak Kotb, UT professor and director of the Mid-South Center for Biodefense and Security.
The National Flu Vaccine Conference was organized in the wake of last winter's vaccine crisis, in which half of the nation's supply was discarded for fear of contamination. Only two companies still make flu vaccine for the American market, down from a dozen ten years ago.
When Britain's Chiron Corp. was barred from shipping vaccine into the country the only other source was Aventis.

Holland's Answer To Genentech?
The West Nile virus might have killed fewer Americans than lightning
strikes, but it's certainly gotten a lot more publicity.
That's good news for Crucell. (CRXL) The Dutch biotech announced this month
that an antibody it derived from West Nile survivors showed good results in
animal studies.
This came on top of its earlier success with a West Nile vaccine that begins
human trials later this year.
Even if the human trials go well, it will be a few years before such
treatments go on the market.
The first Crucell product Americans are likely to see, in fact, will be an
Ebola vaccine that the U.S. government put on a fast track for approval out
of fear of bioterrorism.
"We have been very successful (at proving) that our (Ebola) vaccine protects
100% in animal studies," said Chief Financial Officer Leon Kruimer. "We're
also aiming to get that vaccine for safety testing in man this year."
Meanwhile, Crucell isn't lacking for revenue. It owns a valuable "cell
line" - a family of self-replicating human cells drawn from fetal tissue
that it licenses for genetic research.
Current licensees included Sanofi Aventis, (SNY) Chiron, (CHIR)
GlaxoSmithKline (GSK) and Merck. (MRK)
Kruimer says Crucell's cell line is unique.
"Other cell lines were based traditionally on tumergenic tissue," he said.
"(Our) cell line is the only one in the world based on entirely healthy
tissue."
THE FINANCIALS
Although its revenue has grown rapidly in the last year, Crucell continues
to operate at a loss.
In the first quarter it lost 18 cents a share, narrowing a 23-cent shortfall
the prior year. Revenue gained 57% to $7.7 million.
First Call projections were not available. Securities Associates, an
investment firm that follows Crucell, expects revenue this year to grow 20%
to $35 million.
THE COMPANY
In its present form, Crucell came about through a 2000 merger of IntroGene
with U-Bisys.
IntroGene specialized in gene therapy and owned the cell line.
U-Bisys had an antibody technology called MAbstract, which helps find
antibodies for things like the West Nile vaccine.
Last year Crucell brought a third firm into the mix, a tiny biotech called
Chromegenics.
Its technology, still in development, might help boost production of
antibodies for research.
"All companies that make proteins suffer from low yields," Kruimer said.
"It's a well-known problem."
LOOKING AHEAD
Despite its history of buyouts, Kruimer says Crucell plans to stick to its
core focus on vaccines and infectious diseases. Any acquisitions would be
opportunistic.
In a research note, Securities Associates analysts said Crucell's technology
could have a big impact on the biotech field.
They call the firm "Holland's answer to Genentech." (DNA)
"The value of Crucell lies in the leverage potential of its unique
technology across a broad range of products," analysts wrote. "This company
is unlikely to reach profitability for another two years but, when it does,
we believe the earnings potential could be substantial."

- Ebola: Auftrag von US Behoerden
- West Nile Vaccin
- Malaria
- TBC - http://biz.yahoo.com/pz/050908/85546.html
Also jetzt dann grosse Interesse (d.h.
Crucell versucht Take-over von Schweizerische "Berna Biotech".
Also Berna bietet Crucell "vertical Integration".
Crucell and Berna Biotech: Together Creating the Leading Independent Vaccine Company
Leiden/Berne, 1 December 2005: Crucell N.V. ("Crucell"), a Dutch biotechnology company focused on developing products to prevent and treat infectious diseases, and Berna Biotech A.G. ("Berna"), an established Swiss vaccines company, jointly announced today that Crucell intends to launch a recommended all share offer for the entire issued share capital of Berna at an exchange ratio of 0.447 shares of Crucell for each Berna share. The offer values each Berna share at CHF 15.72 and equals a total Berna equity value of approximately CHF 591 million (€381 million). The combined company will continue under the Crucell name and will be domiciled in The Netherlands. In addition to its Euronext and Nasdaq listing, the combined Crucell-Berna entity will also be listed on the SWX Swiss Exchange.
Vorlaeufig mal abwarten...
